use 'You and I" always; "You and me" never. the latter is incorrect.
I is the subject, Me is the object.
think of it this way: if you say "you and me are going to the store," but later decide that only YOU are going to the store, would you say "me am going to the store"? no, you'd say "I am going to the store." Therefore, "you and I are going to the store."
